Here’s the mechanism in plain English. NNMT’s job is to “tag” (methylate) nicotinamide — a form of vitamin B3 — using the body’s universal methyl donor, SAM (S-adenosylmethionine). When NNMT is over-active in fat cells, two things happen that researchers care about:

In lab studies, NNMT inhibition lowered 1-methylnicotinamide, raised intracellular NAD+ and SAM, and suppressed lipogenesis in fat cells. In short: it’s a metabolic “efficiency” play, not an appetite drug.

  • Diet-induced obese mice: in the landmark study, systemic NNMT inhibitors reduced white-fat mass and body weight and shrank fat cells, lowered cholesterol, and — notably — did not reduce food intake or cause observed side effects. One quinoline-derived inhibitor cut body weight by more than 10% and abdominal fat by over 30% in mice.
  • Fat cells (3T3-L1 adipocytes): 5-Amino-1MQ treatment reduced lipid accumulation, increased oxygen consumption, and raised cellular NAD+.
  • Later studies: NNMT inhibition combined with diet change further reduced body weight, fat mass, and liver fat, and altered gut-microbiome composition.
